Is it possible to define wich Nic to use for vDR if appliance is having multiple vNIC of different segments?

Do you mean the vNIC or the physical NIC?
Usually VDR has a single vNIC (apart VDR2 that has 2 vNIC)
ESXi which is hosting vDR has 2 NIC which are connected to different LAN segment. For vDR will also has assigned 2 different vNIC from one from each NIC. It is possible to divert backup traffic to vNIC 2 if vDR is registered using vNIC 1?
