I have a cluster of 3 ESXi 5.0 host with Center 5.5 and need to upgrade the system to 6.5.
If I have licensed for ESXi 5.0 and Center 5.5 and then upgrade to 5.5 & 6.0 respectively in the process of upgrade to 6.5.
Should I need to buy new licenses or the old licenses will be updated with the upgrade.????

version 5.x, and 6.x require different license keys.
Whether you need to purchase new licenses depends on whether you have an active subscription (SnS), or had one when the version you are going to upgrade to went GA (General Availability). In case you have/had an active SnS, you should be able to upgrade the license keys in your My VMware portal for free.
If SnS has expired, you may either reinstate, or purchase new licenses. The option that's better (less expensive) for you, depends on when SnS has expired.

You would need new licenses for 6.x version - https://docs.vmware.com/en/VMware-vSphere/6.0/vsphere-esxi-vcenter-server-602-upgrade-guide.pdf - Page 223 onward.

Yes you required new license key even though it will upgrade as 60 days evaluation period
