Let 's say we have three DC in three different states : AZ-DC, CA-DC, NY-DC.
1) Can we have a single Work load domain that extends to all three above mentioned DC?
2) Let say we have a single VCF region with three work load domains : WLD1,WLD2, WLD3, can we use the single NSX instance from management domain for these three work load domains?

You can do multi-location workload domains as of version 4.2. To do this, I believe you need 10MBs of bandwidth and a 50 millisecond latency. I am not sure of any the other requirements. I plan on the possibility of doing this at some of our smaller locations in the future. For now, we just did two separate instances. One at our production site and one at or DR site.

there are multiple topology for the VCF deployment, especialy in case of multiple locations or multiple availability zones or regions. Also another factor should be taken into consideration which is the Single Instance of VCF or Multiple Instances.
